چکیده

Sarcomas In mice were induced by i.m. and s.c. administra tion of 63Niand 36S-labeled nickel subsuifide (Ni3S2), and the fate of the Ni3S2 was studied in tumors and normal tissues during carcinogenesis. Whole-body autoradiography showed a gradual loss of solubilized 63Ni and 35S radioactivity from the site of injection. There was also a loss of nonsolubilized dust particles which appeared to be phagocytized by reticuloendothelial cells in the liver, spleen, and regional lymph nodes. Microautoradiography showed that the totally dominating ra dioactivity within both the 63Ni3S2and the Ni335S2-induced tumors was associated with dust particles. There was no spe cific or excessive localization of solubilized radioactivity in the tumors or in métastases (when present). Two patterns of lo calization of dust particles within the tumors were observed: one with particles concentrated in a central part of the tumor and one with the particles present in the periphery of the tumor. X-ray powder diffraction of the insoluble crystalline material in the tumors indicated that a conversion of the aNi3S2 to aNi7S6 and /JNiS had occurred.

Recent research on nickel carcinogenesis.

Research on nickel carcinogenesis from 1975 to March 1980 is reviewed. Epidemiological studies have strengthened the evidence that workers in nickel refineries have increased risks of cancers of the nasal cavities and lungs. Clinical investigations have resulted in improved diagnosis, classification, and management of cancers of respiratory organs in nickel refinery workers. Carcinogenicity tes...

Magnesium inhibits nickel-induced genotoxicity and formation of reactive oxygen.

Nickel compounds are recognized to cause nasal and lung cancers. Magnesium is an effective protector against nickel-induced carcinogenesis in vivo, although its mechanisms of protection remain elusive. The effects of magnesium carbonate on the cytotoxicity and genotoxicity induced by nickel subsulfide were examined with respect to the inhibition of cell proliferation, micronuclei formation, DNA...

Methylation of RAR-β2, RASSF1A, and CDKN2A genes induced by nickel subsulfide and nickel-carcinogenesis in rats.

OBJECTIVE To investigate the expression variation of RAR-β2, RASSF1A, and CDKN2A gene in the process of nickel-induced carcinogenesis. METHODS Nickel subsulfide (Ni(3)S(2)) at dose of 10 mg was given to Wistar rats by intramuscular injection. The mRNA expression of the three genes in induced tumors and their lung metastasis were examined by Real-time PCR. The methylation status of the 5' regi...

Lifetime effects of intratracheally instilled nickel subsulfide on B6C3F1 mice.

A nickel subsulfide (Ni3S2) lung tumor model for the study of metal carcinogenesis was evaluated using intratracheally dosed B6C3F1 mice. A preliminary study of the survival of mice 14 days after a single intratracheal dose of Ni3S2 displayed an LD50 of 4 mg/kg. A lifetime study was then initiated using five graded doses of Ni3S2 or saline alone, administered once a week for 4 weeks. Animals wh...
